マーカー発現との相関について

要旨

Cyclooxygenase type-2 (COX-2) is overexpressed in malignant tumors including ovarian cancer, although the mechanismof up-regulation is unclear. The present study aimed to measure the level of COX-2 expression in serous epithelialovarian carcinomas in comparison with adenomas, and also to investigate the relationship between COX-2 expression,8-hydroxy-2'-deoxyguanosine (8-OHdG), cellular proliferation (Ki-67 expression), apoptotic scores and the degree oftumour infiltrating lymphocytes (TIL). The levels of COX-2, 8-OHdG and Ki-67 expression were determined by immunohistochemistryon paraffin tissue sections of serous ovarian adenocarcinoma (n = 30, stage I 9 cases, stage II 1 case,stage III 14 cases, and stage IV 6 cases), and of adenoma (n = 14). COX-2 expression in serous adenocarcinomas (90%)was significantly greater than that in adenomas (57.1%) (p < 0.05). There was also a significant difference in the 8-OHdGexpression levels (carcinoma 90% vs adenoma 14.3%, p < 0.01) and Ki-67 positive cells (carcinoma 16.3 ± 13.9%,adenoma 2.4 ± 2.2%, p = 0.0006) but there was no significant difference in the apoptotic scores (carcinoma 16.6 ± 15.6%adenoma 6.4 ± 4.8%).We observed significant correlations between a poor prognoses and strong expression of COX-2, increased numbers ofKi-67, and lower apoptotic scores but no significant correlation between 8-OHdG expression. We also observed asignificant positive correlation between COX-2 and 8-OHdG expression levels and a negative correlation between COX-2 expression and the level of TIL. Our results suggest that tissue oxidative stress may affect COX-2 expression resultingin immune suppression and a poor prognosis in ovarian carcinoma patients.
